I had a back fence with 158' of blackberries; when I discovered rats running along the fence and living in the brambles, I used a product called Brush-B-Gone. We sprayed it on the leaves; and in 24 hours the leaves began to wither and in about a week, the stems shriveled and turned yellow and died. However, any plant that the spectracide gets on--will ultimately die. So you have to follow the instructions to the letter and not spray on a windy day or when you are expecting imminent rain-it has to have a chance to work systemically. Then in a couple of weeks, I'd give'm another shot in case the root didn't completely die off. Good luck!

Don't try and kill the brambles now. Pick the berries! It's the best time of year for that and the berries are delicious with ice cream or yogurt, or make some jam!
Then, after the season is over, cut them right back as suggested in this thread, and use the weedkiller on the cut stems.

Roundup, Bayer and Ortho all produce similar results. If you want to step it up go to a brush killer vs. weed killer. For example I needed to kill some poison ivy (and weeds) and that stuff just rocked the hell out of the weeds. They turned brown within hours and died the next day. This particular product was Roundup Poison Ivy Killer.

The best thing for brambles is sbk brushwood killer it's espcially for woody weeds and is inactive once it hits the soil so wont harm any other plants.

There are no weed killer for brambles...weed killer only kill weeds. You will have to dig it up with a pick axe.

Use a good strong weed killer and clear the area when the weeds are dead then you can cover the area with a sheet of polythene then cover with pea shingle or similar this will help prevent the weeds growing back.

All weedkillers are made up of one agent - SODIUM CHLORATE - go and buy a tub of sodium chlorate, just that. Don't go for any brands because they all contain this agent in varying degrees. Sodium chlorate kills everything. Sprinkle it over the brambles and the leaves will die off first, then tackle the roots. One 500g tub by Doff costs £1.25 at my local £1-shop.

Don't use gasoline or anti-freeze, those things will make the soil toxic. Visit your local handyman store and ask them there. I have used roundup for many things, I don't know why it isn't working for you, but cut the bramble down to a stalk and then spray directly into the stalk wound.

Roundup is not strong enough use "Brush Clear" I think that's the name but it could be a derivation, If you go to a good garden centre they will have the right stuff, and possibly many brands, check the back of the label or simply ask a staff member

As per Charlotte M advice, you need a product containing MCPA 24D-T it is sold in the UK as SPK Brushwood Killer. Please be very careful with it as it is a very nasty product and will kill fish in ponds nearby and other wildlife.

Elbow grease! Pull up the smaller canes directly out of the soil to disrupt their root systems, then dig out the rest, having cut them short to dispose of the top growth without losing an eye. Pulling on the mature canes will usually also lift out any rooting runners, saving you some digging.
I never use weedkillers if I can avoid them, preferring to garden organically.
